Targeting oncogenic signaling pathways in head and neck cancer using nano-phytomedicine: Molecular insights and therapeutic potential

Abstract

Head and neck squamous cell carcinoma (HNSCC) is a highly destructive cancer known for its molecular heterogeneity and resistance to standard treatments, necessitating the development of new therapeutic options. Numerous carcinogenic signaling pathways, including EGFR/MAPK, PI3K/Akt/mTOR, NF-κB, STAT3, and Wnt/β-Catenin, are intrinsically associated with the pathophysiology of HNSCC, regulating cell proliferation, survival, and metastasis. This review demonstrates the therapeutic use of plant-derived phytomedicines to target interrelated clinical processes. It investigates the molecular mechanisms through which compounds such as genistein, EGCG, curcumin, quercetin, resveratrol, and apigenin concurrently influence multiple oncogenic pathways, resulting in their diverse anti-cancer effects. Nano-phytomedicine combines the delivery efficiency of nanotechnology with the anticancer potential of bioactive phytocompounds, emerging as a promising treatment. By altering key oncogenic signaling pathways, promoting apoptosis, preventing epithelial-mesenchymal transition, and reducing cancer stemness, various therapeutic strategies can be used to treat cancer. Despite their potential, the rapid systemic clearance, low bioavailability, and poor solubility of these substances have inhibited their clinical translation. Nano-phytomedicine can help overcome these pharmacokinetic limitations. Encapsulating potent natural agents within advanced nanocarriers can enhance stability, prolong drug release, and improve tumor-specific accumulation. This review critically analyzes the use of nano-formulated phytocompounds to selectively target dysregulated oncogenic signaling pathways in the pathogenesis of head and neck cancer. Recent advancements in nanoparticle delivery methods focus on molecular interactions with signaling networks and their therapeutic potential. This review emphasizes nano-phytomedicine as a transformative strategy for precision-targeted treatment in head and neck cancer and proposes potential approaches for clinical application. This approach minimizes off-target effects and enhances the effectiveness of existing HNSCC treatments. Additionally, this review demonstrates the potential of nano-phytomedicine as a promising preclinical strategy for the treatment of head and neck cancer, necessitating further clinical validation.
